Increased expression of long non-coding RNA TUG1 associates with poor prognosis of hepatocellular carcinoma

چکیده
Background: Previous studies found that lncRNA TUG1 expression was up-regulated in hepatocellular carcinoma (HCC) tissues. In the present study, we aimed to investigate the clinical significance and prognostic value of lncRNA TUG1 in HCC. Methods: Matched fresh specimens of HCC and paracarcinomatous liver tissues were obtained from 137 patients who underwent hepatic resection. qRT-PCR assay was performed to determine the expression level of lncRNA TUG1. Kaplan-Meier survival curves and Cox proportional hazards regression analysis were performed using SPSS software. Results: lncRNA TUG1 expression was significantly higher in HCC tissues compared with normal adjacent liver tissues (P<0.001). High lncRNA TUG1 expression level was observed to be closely correlated with liver cirrhosis (P=0.015), Child-Pugh score (P=0.004), tumor size (P=0.016), tumor number (P=0.001), TNM stage (P<0.001), Edmondson-Steiner grade (P<0.001), and vein invasion (P<0.001). A significant relationship was found between lncRNA TUG1 expression and 5-year overall survival (P=0.009, log-rank test). Furthermore, in a multivariate Cox model, we found that lncRNA TUG1 expression was an independent poor prognostic factor for 5-year overall survival in HCC (HR=2.966, CI=1.273-9.783, P=0.022). Conclusion: This study demonstrated that increased lncRNA TUG1 expression levels were correlated with poor prognosis of patients with HCC, indicating that lncRNA TUG1 may serve as a novel prognostic marker for HCC after hepatic resection.

The prognostic potential and carcinogenesis of long non-coding RNA TUG1 in human cholangiocarcinoma
Cholangiocarcinoma (CCA) is a fatal disease with increasing worldwide incidence and is characterized by poor prognosis due to its poor response to conventional chemotherapy or radiotherapy. Long non-coding RNAs (lncRNAs) play key roles in multiple human cancers, including CCA. Cancer progression related lncRNA taurine-up-regulated gene 1 (TUG1) was reported to be involved in human carcinomas. H...

OBJECTIVE The long non-coding RNAs (lncRNAs) study has gradually become one of the hot topics in the field of RNA biology. However, little is known about the pathological role of lncRNA TUG1 in clear cell renal cell carcinoma (ccRCC) patients. This study attempted to investigate the association of lncRNA TUG1 expression with progression and prognosis in ccRCC patients. PATIENTS AND METHODS Us...
متن کاملLong Non-Coding RNA TUG1 Expression Is Associated with Different Subtypes in Human Breast Cancer
Taurine upregulated 1 gene (TUG1) is a long non-coding RNA associated with several types of cancer. Recently, differential expression of TUG1 was found in cancerous breast tissues and associated with breast cancer malignancy features. Although this is evidence of a potential role in breast cancer, TUG1 expression could not be associated with different subtypes, possibly due to the small number ...

متن کاملTUG1: a pivotal oncogenic long non-coding RNA of human cancers.
Long non-coding RNAs (lncRNAs) are a group greater than 200 nucleotides in length. An increasing number of studies has shown that lncRNAs play important roles in diverse cellular processes, including proliferation, differentiation, apoptosis, invasion and chromatin remodelling. In this regard, deregulation of lncRNAs has been documented in human cancers. TUG1 is a recently identified oncogenic ...
